Cortisol is a steroid hormone and belongs to the glucocorticoid class. When used as a drug, it is called hydrocortisone. It is produced in many animals, mainly in the fascia area of the adrenal cortex. The quantity it generates in other organizations is relatively small. Its release has a day-night cycle, and its release increases under conditions of stress and low blood sugar concentration. Its function is to increase blood sugar through glucose production, suppress the immune system, and assist in the metabolism of fats, proteins and carbohydrates. It also reduces bone formation. Cortisol stimulates glucose production (the synthesis of "new" glucose from non-carbohydrate sources, mainly in the liver, but in some cases also in the kidneys and small intestine). Its net effect is an increase in the concentration of glucose in the blood, which is further supplemented by a decrease in the insulin sensitivity of peripheral tissues, thereby preventing the tissues from obtaining glucose from the blood.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 20 10 5 2.5 1.25 0.625 0.312 0
OD 0.19 0.24 0.31 0.55 0.97 1.38 1.96 2.52

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human SC spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 81-101 96
Plasma(n=8) 92-105 101

| Linearity

Samples were spiked with high concentrations of Human SC and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4)
1 : 2 Range (%) 83-95 88-96
Average (%) 91 93
1 : 4 Range (%) 89-104 87-108
Average (%) 93 98
